Ozone Therapy Protects Against Rejection in a Lung Transplantation Model: A New Treatment?

No satisfactory treatment exists for chronic rejection (CR) after lung transplantation (LT). Our objective was to assess whether ozone (O<sub>3</sub>) treatment could ameliorate CR. Male Sprague-Dawley inbred rats (n&#xa0;= 36) were randomly assigned into four groups: (1) control (n&#xa0;= 6), (2) sham (n&#xa0;= 6), (3) LT (n&#xa0;= 12), and (4) O<sub>3</sub>-LT (n&#xa0;= 12). Animals underwent left LT. O<sub>3</sub> was rectally administered daily for 2 weeks before LT (from 20 to 50 &#x3bc;g) and 3 times/wk (50 &#x3bc;g/dose) up to 3 months. CR; acute rejection; and Hspb27, Prdx, Epas1, Gpx3, Vegfa, Sftpa1, Sftpb, Plvap, Klf2, Cldn5, Thbd, Dsip, Fmo2, and Sepp1 mRNA gene expression were determined. Severe CR was observed in all animals of LT group, but none of the O<sub>3</sub>-LT animals showed signs of CR, just a mild acute rejection was observed in 1 animal. A significant decrease of Hspb27, Prdx, Epas1, Gpx3, Vegfa, Sftpa1, Sftpb, Plvap, Klf2, Cldn5, Thbd, Dsip, and&#xa0;Fmo2 gene expression in the O<sub>3</sub>-LT group was observed CONCLUSIONS: O<sub>3</sub> therapy significantly delayed the onset of CR regulating the expression of genes involved in its&#xa0;pathogenesis. No known immunosuppressive therapy has been capable of achieving similar results. From a translational point of view, O<sub>3</sub> therapy could become a new adjuvant treatment for CR in patients undergoing LT.

Previous studies have shown that humoral, endogenous and somnogenic, delta sleep-inducing peptide (DSIP) has influence on insomnia, pain, adaptation to stress, epilepsy, etc. We investigated the potential of DSIP and its analogue DSIP-12 (a nonapeptide with alanine in position 2 of DSIP molecule substituted by beta-alanine) to antagonize metaphit (1-[1(3-isothiocyanatophenyl)-cyclohexyl]piperidine) induced generalized, reflex audiogenic seizures in adult male Wistar albino rats. The rats divided in four groups received (i.p.): saline; metaphit; metaphit+DSIP; and metaphit+DSIP-12, respectively. Metaphit-treated animals displaying seizure in eight previous tests received DSIP or DSIP-12 and afterwards audiogenic stimuli were applied at hourly intervals for the next 30 h. The animals were exposed to sound stimulation 60 min after metaphit administration and further on at hourly intervals. Incidence and severity of seizures were behaviorally analyzed. Selected EEGs and power spectra were recorded and analyzed. Metaphit led to hypersynchronous epileptiform activity (polyspikes and spike-wave complexes) and increased power spectra 0.5-30 h after the treatment. Severity of metaphit seizures increased with time to reach the peak 7-12 h after injection. DSIP and DSIP-12 significantly (*P&lt;0.05 and **P&lt;0.01) increased in delta and theta frequency bands and decreased the incidence, mean seizure grade and duration of metaphit convulsions. The results suggest that DSIP and DSIP-12 may be considered as potential antiepileptics in the animal model, DSIP-12 being more efficient than DSIP.

Expression and Purification of Delta Sleep-Inducing Peptide Fused with Protein Transduction Domain and Human Serum Albumin in Pichia pastoris.

Sleep is a natural part of every individual's life. Delta sleep-inducing peptide (DSIP) is a nonapeptide that could promote sleep through the induction of slow wave sleep. However, little is known about the pharmacological effect of DSIP on insomnia. The main objective of this study was to analyze the pharmacological effect of DSIP on insomnia. We designed a fusion protein containing N-terminal TAT-based transduction domain followed by human serum albumin and DSIP and designated this protein as PHD fusion protein. The PHD fusion protein were expressed in Pichia pastoris and purified. Mice were administered single subcutaneous injections three concentrations of PHD fusion protein (0.5, 1, 2 mg/kg), and the pharmacological activity of PHD fusion protein was studied using classic pentobarbitalinduced sleep test. We expressed the PHD fusion protein in P. pastoris; furthermore, the PHD fused protein was purified to near homogeneity by DEAE Sepharose FF, Phenyl Sepharose HP and Blue Sepharose 6 FF. Our result showed that the increase of pentobarbital-induced hypnotic effect characterized by reducing sleep latency and prolonged sleep duration was observed for increasing concentrations of PHD fusion protein (P&lt;0.05); moreover, different dose of PHD fusion protein could induce the mice to re-sleep in a dose-dependent manner, whereas higher doses of PHD fusion protein (1.0, 2.0 mg/kg) significantly increased the rate of sleep re-onset compared with the vehicle group of mice (P&lt;0.05). PHD fusion protein increased the hypnotic effects of pentobarbital by reducing sleep latency and prolonged sleep duration. The present study suggested PHD fusion protein could be a new drug candidate for insomnia.

[Effects of the &#x3b4;-sleep inducing peptide on neuronal activity in the dorsal hippocampus of rats with different behavior in the open field after stimulation of the positive and negative emotionalistic structures of the brain].

We studied the cerebral mechanisms of positive and negative emotions in rats with different behavior in open field, reflecting stress resistance and neuronal effects of delta-sleep-inducing peptide (DSIP). In 20 male Wistar rats 107 neurons of dorsal hippocampus (57 neurons in active in open field--prognostically resistant to emotional stress and 50 inpassive--prognostically predisposed rats) were registered after positive (lateral hypothalamus--LH) and negative (ventromedial hypothalamus--VMH) emotional centers electric stimulation. Hippocampal neurons in active rats were less sensitive to stimulation of LH and VMH compared with passive ones. DSIP microiontophoretic application before LH stimulation decreases neuronal responses in both active and passive animals. DSIP increases dorsal hippocampus neurons sensitivity to VMH stimulation in active rats and decreases in passive ones.

[Endogenous anticonvulsants: neuropeptide Y and delta sleep inducing peptide].

The same neuropeptides regulate both cicle sleep-wake and excitability of the brain. Literature data together with our results support the idea that delta sleep--inducing peptide and neuropeptide Y could represent one of the factors of the endogenous stabilization of brain excitability and potent antiepileptic in generalized metaphit-induced audiogenic convulsive activity. The same holds true for DSIP analogues.

Structural-functional organization of neurons in the cerebral cortex of rats with different levels of resistance to emotional stress in conditions of exposure to delta sleep-inducing peptide.

Wistar rats with different levels of resistance to emotional stress (ES) were subjected to stress and brain sections stained with Nissl cresyl violet were used for quantitative analysis of the structural organization of neurons in layer V of the sensorimotor cortex. Some animals received delta sleep-inducing peptide (DSIP) 1 h before stress. Control ES-susceptible rats, as compared with resistant rats, had lower levels of normochromic and moderately hypochromic neurons. Normochromic neurons were not seen after stress. Rats susceptible to ES showed particularly sharp decreases in moderately hypo- and hyperchromic neurons, along with increases in the proportions of extremely hypo- and hyperchromic neurons, ghost cells, and ischemically altered cells. After administration of DSIP before stress, ischemically altered cells were not seen in any group: the level of reduction of extremely hyperchromic neurons was smaller in ES-susceptible rats than in ES-resistant rats. It is suggested that brain hypoxia plays a particular role in disorganizing the cortex in conditions of ES, while DSIP has both antistress and antihypoxic properties.

Subcutaneous injections of exogenous delta sleep-inducing peptide in a dose of 100 &#x3bc;g/kg (monthly, 5-day courses) to rats of various age groups (2-24 months) were followed by an increase in the expression of genes for SOD 1 (Sod1) and glutathione peroxidase 1 (Gpx1) in the brain and nucleated blood cells. The expression of these genes was shown to decrease during physiological aging of the body.

In a rat study, a drug that blocks NMDA receptors (dizocilpine) stopped a sleep‑related peptide (delta sleep‑inducing peptide, DSIP) from lowering a brain activity marker (c‑fos) in a specific hypothalamic region. This shows DSIP works through NMDA pathways, but the work is purely basic science and doesn’t give any practical advice for using DSIP in people.
